Typical DosageStandard: 300-600 mg daily of extract standardized to 50% bacosides (e.g., Bacognize or Synapsa). Take with fat for absorption. Allow 8-12 weeks for full effects. Evening dosing preferred due to mild sedation.Standard: 10-20 mg daily. Often combined with CoQ10 (100-300 mg) for synergistic mitochondrial support. Higher doses (40 mg) are used in some research settings.
AdministrationOral (capsules, powder). Fat-soluble — take with a meal containing fat.Oral (capsules, softgels). Best absorbed on an empty stomach. BioPQQ is the most studied branded form.

Mechanism of Action

Bacopa Monnieri

Bacopa's bacosides (bacosides A and B, bacopaside I-VII) enhance synaptic communication by increasing dendritic branching, spine density, and synaptic activity in the hippocampus via modulation of neural cell adhesion molecules (NCAM) and FGF-2. They modulate serotonin through 5-HT3 receptor antagonism (reducing anxiety) and 5-HT2A modulation, dopamine through D1/D2 receptor modulation, and acetylcholine through enhancement of choline acetyltransferase. Bacosides upregulate tryptophan hydroxylase (TPH) and serotonin transporter (SERT) expression, increasing serotonin synthesis and reuptake. The antioxidant properties of bacosides reduce lipid peroxidation and protein carbonylation in the hippocampus via free radical scavenging, protecting neurons from oxidative damage during memory formation. They may enhance CREB phosphorylation and BDNF expression.

PQQ

PQQ activates PGC-1alpha (peroxisome proliferator-activated receptor gamma coactivator 1-alpha), the master transcriptional regulator of mitochondrial biogenesis. PGC-1alpha coactivates NRF-1 and NRF-2, which drive expression of mitochondrial transcription factor A (TFAM) and nuclear-encoded mitochondrial genes—the process of creating new mitochondria in existing cells. This is unique among commercially available supplements. PQQ also provides antioxidant protection through extremely efficient redox cycling at the N5 position; it can undergo thousands of oxidation-reduction cycles before being exhausted, estimated at 5,000x the efficiency of vitamin C. PQQ activates the CREB (cAMP response element-binding protein) signaling pathway and may enhance NGF signaling, supporting BDNF expression, synaptic plasticity, and neuronal survival.
